The confocal microscopy for assessement of transdermal transport of the photosensitizer chlorin E6 after non-ablative photothermolysis

Abstract

Transdermal delivery of the drug using laser nonablative photothermolysis of the epidermis has the prospect of clinical application, but requires further study of the specifics of the drug penetration into all layers of the skin and subcutaneous space. In our work, we demonstrate a method for determining the drug Radagel containing chlorin E6 in the skin layers using confocal microscopy and light energy in the spectrum, which allows us to initiate fluorescence and record its intensity. Nonablative photothermolysis provided a pronounced increase in the fluorescence of chlorin E6 in the deep layers of the dermis, and quantitative determination of the fluorescence intensity allowed us to calculate the coefficient of transdermal transport of the drug, which increased by 27 times after photothermolysis.
